Active antithrombin glycoforms are selectively physiosorbed on plasma extracellular vesicles

چکیده
Antithrombin (AT) is a glycoprotein produced by the liver and principal antagonist of active clotting proteases. A deficit in AT function leads to qualitative deficiency, challenging diagnose. Here we report that may travel physiosorbed on surface plasma extracellular vesicles (EVs), contributing form “EV-protein corona.” The corona enriched specific glycoforms, thus suggesting glycosylation play key role partitioning between EVs plasma. Differences glycoform composition separated from healthy deficiency-affected subjects were also noticed. This suggests deconstructing into its nanostructured components, as EVs, could suggest novel directions unravel pathophysiological mechanisms.

Biologically active Fas antigen and its cognate ligand are expressed on plasma membrane-derived extracellular vesicles.
Exfoliation of plasma membrane components is a directed process that consumes energy and requires active cell metabolism. Proteins involved in regulating the survival and proliferation of eukaryotic cells are released on exfoliated vesicles. We examine here whether the Fas receptor and its cognate ligand (FasL) are present on vesicles shed from high metastatic potential CX-1 cells and low metas...
متن کاملActive endocannabinoids are secreted on extracellular membrane vesicles.
Endocannabinoids primarily influence neuronal synaptic communication within the nervous system. To exert their function, endocannabinoids need to travel across the intercellular space. However, how hydrophobic endocannabinoids cross cell membranes and move extracellularly remains an unresolved problem. Here, we show that endocannabinoids are secreted through extracellular membrane vesicles prod...
متن کاملExtracellular vesicles in the circulation: are erythrocyte microvesicles a confounder in the plasma haemoglobin assay?
Blood contains a mixture of extracellular vesicles from different cell types, primarily platelets, endothelial cells, leucocytes and erythrocytes. Erythrocytes are the most abundant cell type in blood and could, especially in certain pathologies, represent an important source of vesicles. Since erythrocytes contain the haemoglobin components iron and haem, which are potentially toxic, it is imp...

Extracellular vesicles (EVs) such as exosomes and microvesicles mediate intercellular communication and regulate a diverse range of crucial biological processes. Host cells that are damaged, infected or transformed release biomarker-containing EVs into the peripheral circulation, where they can be readily accessed for use in diagnostic or prognostic testing. However, current methods of EV isola...
متن کاملEXTRACELLULAR VESICLES AND THEIR THERAPEUTIC APPLICATIONS: A REVIEW
Background & Aims: Extracellular vesicles are double phospholipid layer vesicles releasing from several cells. These particles contain bio-molecules including mRNA, miRNA, DNA, proteins, and lipids which play a pivotal role in the cell to cell communication. Once delivered, they participate to change target cell function, fate, morphology, differentiation, and growth. The present study is a re...
